Viscocapillary instability in cellular spheroids

چکیده

We describe a viscocapillary instability that can perturb the spherical symmetry of cellular aggregates in culture, also called multicellular spheroids. In condition where cells constituting spheroid get their necessary metabolites from immediate, outer microenvironment, permanent cell flow exists within its rim divide toward core they die. A perturbation induces viscous shear stresses tissue destabilise aggregate. The proposed is nature and does not rely on external heterogeneities, such as pre-existing pattern blood vessels or presence substrate which exert pulling forces. It arises for sufficiently large cell-cell adhesion strengths, cell-renewal rates, metabolite supplies, described by our model parameters. Since spheroids culture are good systems small, avascular tumours, mimicking concentration gradients found vivo, we speculate description applies to microtumour instabilities cancer progression.

The Convective Instability of Boundary-layer Flows over Rotating Spheroids

The continuous development of spinning projectiles and other industrial applications has led to the need to understand the laminar boundary-layer flow and subsequent onset of transition over the general family of rotating spheroids. We begin by finding the laminar boundary-layer flow over a general spheroid. In particular, we distinguish between prolate and oblate spheroids and use an appropria...

Cusp instability in cellular growth

During directional solidification of alloys, a cellular structure can be observed above a critical pulling speed. Under certain conditions, the cusps behind the cells can present a secondary instability characterized by a periodic emission of liquid droplets. These droplets are transported by the solid, but also migrate towards the solid-liquid interface by temperature gradient zone migration. ...

Mechanical Stress Impairs Mitosis Progression in Multi-Cellular Tumor Spheroids

Growing solid tumors are subjected to mechanical stress that influences their growth rate and development. However, little is known about its effects on tumor cell biology. To explore this issue, we investigated the impact of mechanical confinement on cell proliferation in MultiCellular Tumor Spheroids (MCTS), a 3D culture model that recapitulates the microenvironment, proliferative gradient, a...

Proliferation and viability in cellular spheroids of human origin.

The capacity of cells to form growing tumor-like colonies in culture was tested by a new and unlaborious suspension technique. Six of nine tested cell lines formed spheroids, five of which started to grow. The growing spheroids reached a maximal size determined by balanced cell gain in a proliferative, superficial layer and cell death, particularly at larger depth. Sections of spheroids of diff...

Incorporating Cellular Metabolism into Growth Models of Multicellular Tumor Spheroids

Many chemotherapeutic strategies have failed due to pronounced diffusion limitations in tumors induced by poor vasculaturization. Diffusion limitations, present in threedimensional tissue, produce nutrient deficient regions that eventually become necrotic in tumors in vivo. Multicellular spheroids are better models of in vivo tumors than monolayer cultures because they mimic the heterogeneous m...
